PconsFold: improved contact predictions improve protein models
Motivation: Recently it has been shown that the quality of protein contact prediction from evolutionary information can be improved significantly if direct and indirect information is separated. Given sufficiently large protein families, the contact predictions contain sufficient information to predict the structure of many protein families. However, since the first studies contact prediction methods have improved. Here, we ask how much the final models are improved if improved contact predictions are used. Results: In a small benchmark of 15 proteins, we show that the TM-scores of top-ranked models are improved by on average 33% using PconsFold compared with the original version of EVfold. In a larger benchmark, we find that the quality is improved with 15â30% when using PconsC in comparison with earlier contact prediction methods. Further, using Rosetta instead of CNS does not significantly improve global model accuracy, but the chemistry of models generated with Rosetta is improved. Availability: PconsFold is a fully automated pipeline for ab initio protein structure prediction based on evolutionary information. PconsFold is based on PconsC contact prediction and uses the Rosetta folding protocol. Due to its modularity, the contact prediction tool can be easily exchanged. Aplicación de herramientas lean manufacturing para la mejora de la productividad en el årea de almacén de la empresa LA CALERA 2021
La presente investigaciĂłn titulada âAplicaciĂłn de herramientas Lean
Manufacturing para la mejora de la productividad en el årea de almacén de la
empresa LA CALERAâ. Primeramente, tuvo como principal objetivo general
determinar cĂłmo la aplicaciĂłn de herramientas Lean Manufacturing mejora la
productividad en el årea de almacén. Por ello la población que se estudió se
considerĂł los siguientes datos tomados y la cantidad de Ăłrdenes despachadas,
durante las 16 semanas, lo cual se tuvo como variable independiente Lean
Manufacturing y la variable dependiente la productividad.
Asimismo, en esta investigaciĂłn el principal problema que presenta es la baja
productividad en el årea almacén, lo que viene ocasionando una ineficiencia e
ineficacia tanto en los operadores y en los procesos internos del årea. Después
de la aplicaciĂłn de las herramientas Lean Manufacturing se logrĂł un aumento en
la productividad en 14.65%, de la eficiencia en 6.01% y de la eficacia en 21.79%
en el ĂĄrea correspondiente, esta investigaciĂłn es de enfoque cuantitativo, tipo
aplicada, y nivel explicativo.
Por ello, se concluye que con la implementaciĂłn de la herramienta Lean
Manufacturing se consiguió incrementar la productividad en el årea de almacén
de la empresa Calera 2021
Structural basis for the interaction of the chaperone Cbp3 with newly synthesized cytochrome b during mitochondrial respiratory chain assembly
Assembly of the mitochondrial respiratory chain requires the coordinated synthesis of mitochondrial and nuclear encoded subunits, redox co-factor acquisition, and correct joining of the subunits to form functional complexes. The conserved Cbp3-Cbp6 chaperone complex binds newly synthesized cytochrome b and supports the ordered acquisition of the heme co-factors. Moreover, it functions as a translational activator by interacting with the mitoribosome. Cbp3 consists of two distinct domains, an N-terminal domain present in mitochondrial Cbp3 homologs, and a highly conserved C-terminal domain comprising a ubiquinol-cytochrome c chaperone region. Here, we solved the crystal structure of this C-terminal domain from a bacterial homolog at 1.4 Ă
resolution, revealing a unique all-helical fold. This structure allowed mapping of the interaction sites of yeast Cbp3 with Cbp6 and cytochrome b via site-specific photo-crosslinking. We propose that mitochondrial Cbp3 homologs carry an N-terminal extension that positions the conserved C-terminal domain at the ribosomal tunnel exit for an efficient interaction with its substrate, the newly synthesized cytochrome b protein
Untersuchung und EindÀmmung eines SARS-CoV-2-Alpha-Ausbruchs in einer Pflegeeinrichtung im Landkreis Dithmarschen, Juni 2021
Bewohnende und Personal in Pflegeeinrichtungen gehören zu den Personengruppen, die ab dem 27.12.2020 in Deutschland priorisiert die COVID-19-Impfung erhielten. Diese zeigte eine gute Wirksamkeit hinsichtlich des Schutzes vor Infektionen mit der Alpha-Variante und eine sehr hohe Wirksamkeit vor schweren KrankheitsverlĂ€ufen. In einer Pflegeeinrichtung im Landkreis Dithmarschen kam es zwischen dem 01.04. und 23.06.2021 dennoch zu einem SARS-CoV-2-Ausbruch, bei dem eine hohe Anzahl vollstĂ€ndig geimpfter Personen infiziert wurde und teilweise schwer erkrankte oder verstarb. Beschrieben werden zum einen die Methoden, die bei der Ausbruchsuntersuchung im Pflegeheim angewandt wurden, und zum anderen die MaĂnahmen, die schlussendlich zur AusbruchseindĂ€mmung beitrugen.Peer Reviewe

The production of tt⟠, W+bb⟠and W+cc⟠is studied in the forward region of protonâproton collisions collected at a centre-of-mass energy of 8 TeV by the LHCb experiment, corresponding to an integrated luminosity of 1.98±0.02 fbâ1 . The W bosons are reconstructed in the decays WââÎœ , where â denotes muon or electron, while the b and c quarks are reconstructed as jets. All measured cross-sections are in agreement with next-to-leading-order Standard Model predictions.The production of , and is studied in the forward region of proton-proton collisions collected at a centre-of-mass energy of 8 TeV by the LHCb experiment, corresponding to an integrated luminosity of 1.98 0.02 \mbox{fb}^{-1}. Observation of the B0 â Ï0Ï0 decay from an amplitude analysis of B0 â (Ï+Ïâ)(Ï+Ïâ) decays
Protonâproton collision data recorded in 2011 and 2012 by the LHCb experiment, corresponding to an integrated luminosity of 3.0 fbâ1 , are analysed to search for the charmless B0âÏ0Ï0 decay. More than 600 B0â(Ï+Ïâ)(Ï+Ïâ) signal decays are selected and used to perform an amplitude analysis, under the assumption of no CP violation in the decay, from which the B0âÏ0Ï0 decay is observed for the first time with 7.1 standard deviations significance. The fraction of B0âÏ0Ï0 decays yielding a longitudinally polarised final state is measured to be fL=0.745â0.058+0.048(stat)±0.034(syst) . The B0âÏ0Ï0 branching fraction, using the B0âÏKâ(892)0 decay as reference, is also reported as B(B0âÏ0Ï0)=(0.94±0.17(stat)±0.09(syst)±0.06(BF))Ă10â6
A study of CP violation in B-+/- -> DK +/- and B-+/- -> D pi(+/-) decays with D -> (KSK +/-)-K-0 pi(-/+) final states
A first study of CP violation in the decay modes and , where labels a or meson and labels a or meson, is performed. The analysis uses the LHCb data set collected in collisions, corresponding to an integrated luminosity of 3 fb. The analysis is sensitive to the CP-violating CKM phase through seven observables: one charge asymmetry in each of the four modes and three ratios of the charge-integrated yields. The results are consistent with measurements of using other decay modes

Decay candidates with pion pairs that have invariant mass in the range 0.5-1.3 GeV/ and with muon pairs that do not originate from a resonance are considered. The first observation of the decay and the first evidence of the decay are obtained and the branching fractions are measured to be and , where the third uncertainty is due to the branching fraction of the decay , used as a normalisation.A search for the rare decays Bs0âÏ+ÏâÎŒ+ÎŒâ and B0âÏ+ÏâÎŒ+ÎŒâ is performed in a data set corresponding to an integrated luminosity of 3.0 fbâ1 collected by the LHCb detector in protonâproton collisions at centre-of-mass energies of 7 and 8 TeV .
